Collection of sperm samples from birds

Summary of method


Sperm samples (~ 0.5-3 µl) are collected with microcapillary tubes from the cloacal protuberance of male birds after a gentle massage and diluted in a small volume (~20 µl) of Phosphate-buffered saline (PBS) solution before fixed in ~300 µl of 5% formaldehyde solution for later slide preparation.






Sperm samples can be collected from male birds during their breeding periods. the sperm are produce in the testes in male birds, and are transported through the deferent duct for temporary storage at the cloaca, from which they ejaculate. The temporary storage of sperm in the cloaca makes the cloaca enlarge (protuberance). This is key to knowing whether a male bird will have sperm.


Sampling materials


1. Tube containing formalin

2. Tube containing Phosphate Buffered Saline (PBS),

3. Droppers or capilliary holders,

4. Microcapilliary tubes


Procedure for sperm collection

  • Remove the lid/cap of the tube containing formalin

  • Take a drop of PBS from the tube containing PBS and place inside the lid/cap removed from the formalin tube.

  • Take one microcapillary and insert into the dropper/holder

  • Collect the sperm sample from the bird suing the microcapilliary on the holder. Note NOTE: microcapillary suck up the sperm as it make contact with it

  • Gentle discharge the content of the microcapillary into the PBS on the lid from the formalin tube.

  • Gently pick the lid with the content and cover the formalin with it.

  • NOTE: PBS help the sperm cell not be clued together as would normally without it. It also help to cushion the sperm cell from the effect of the formalin.
